The Schmidt Initiative for Long COVID has funded Western University to conduct a Phase III, double-blind, placebo-controlled, multi-arm platform study. FHCSD will enroll participants alongside a consortium of international partners from Brazil, Canada, Italy, Uganda, Zambia, and the United States. This large-scale platform study will evaluate the efficacy of multiple repurposed drug interventions, targeting the underlying mechanisms of Long Covid as determined by LC-Revitalize’s robust proteomic studies in a diverse sample of participants globally. This study aims to develop comprehensive treatment regimens, improving outcomes for individuals living with Long Covid. The first phase, involving 348 participants, will investigate two repurposed drugs, upadacitinib and pirfenidone, over a 3-month 12-week) period, each paired with its own placebo, followed by an interim analysis. After completing the 3-month treatment, the original 348 participants will be monitored for an additional 3 months for symptom rebound and safety. The second phase, guided by the results of the first phase, will be followed by a final analysis. In this phase, the study may continue with one or both repurposed drugs, combine one with an add-on repurposed drug, or introduce completely new repurposed drugs.
